Join Takeda as a Senior Research Associate I, Protein Purification and Characterization where you will produce therapeutics in the areas of oncology, neuroscience and gastrointestinal diseases, especially bispecifics/multispecifics and alternate/non antibody formats using multi column chromatography. You will also characterize purified proteins using suitable analytical techniques to enable isolation and identification of the required format. As part of the, Therapeutic Generation and Screening (TGS) team you will report to the Principal Scientist and support AI/ML approaches to develop the next generation of biotherapeutics, by working to innovate, enhance, and drive automated purification platforms and an IT infrastructure necessary to advance the biotherapeutic pipeline.

How you will contribute:

  • To support Antibody Discovery, designs, purifies, and characterizes complex biotherapeutic molecules using optimal selection of chromatographic resins and binding assays to advance novel therapeutics.
  • Conduct complex protein purification and characterization experiments from small scale to large-scale and initiates trouble shooting steps and automation to increase throughput when appropriate.
  • Perform routine biosensor (SPR/BLI) based biomolecular interactions and data analysis.
  • Exercise technical judgement in the interpretation of results and follow appropriate procedures to record data in internal databases and electronic lab notebooks.
  • Communicate research and presents findings to internal stakeholders at team meetings.
  • Maintain responsibility for laboratory instrumentation and supplies.

    Minimum Requirements/Qualifications:

    • MS in Biochemistry, Chemistry or Molecular Biology (or equivalent) with 1+ year experience.
    • B.S. in Biochemistry, Chemistry or Molecular Biology (or equivalent) with 3+ years of experience.
    • Prior experience developing methods for analytical SEC to enable identification of the test species and for BLI (Octet) or SPR (Biacore) to determine binding affinities and/or binding kinetics is a must.
    • Prior experience executing appropriate purification and characterization methods to purify bispecific/multispecific antibody like molecules, with a deep understanding of how different formats and mutations affect the purification process.
    • Prior experience purifying different formats using automated platforms to support AI/ML approaches to protein design is a plus.
    • Prior experience purifying and refolding proteins from bacteria is a plus.
    • Drive purification strategies including use of appropriate hosts to get the best protein and can guide CRO's using internally developed protocols. protocols.
    • Demonstrate an in-depth knowledge of protein characterization techniques, including CE-SDS, analytical SEC for purity and Octet/Biacore for determination of protein-protein interactions, binding kinetics, affinities, and epitope binning.
    • Recognize problems and consider the significance of irregularities within protein purification and characterization experiments.
    • Demonstrate a basic knowledge of workflows upstream (e.g., protein expression) and downstream (e.g., mass spec, cell based functional assays) of protein purification and how those workflows can affect the purification workflow.

        Base Salary Range: $89,000.00 to $117,000.00, based on candidate professional experience level. Employees may also be eligible for Short-term and Long-Term Incentive benefits as well. Employees are eligible to participate in Medical, Dental, Vision, Life Insurance, 401(k), Charitable Contribution Match, Holidays, Personal Days & Vacation, Tuition Reimbursement Program and Paid Volunteer Time Off. The final salary offered for this position may take into account a number of factors including, but not limited to, location, skills, education, and experience.
